Production and consumption of B group vitamins in situ. ArchiMer
Kurata, A.
The changes in concentration of thiamine, biotin and vitamin B sub(12) in water of the soaked bottles for 6 h in situ in the north basin of Lake Biwa were examined successively over 30 hours. The concentrations of the vitamins changed comparatively during 6 h. soaking in situ. Generally, the concentrations of thiamine, biotin and vitamin B sub(12) were in high levels in the morning and in low levels in evening both in the soaked bottles and in the surrounding water. The fluctuation ranges in concentration of vitamins in a 5 m deep layer were greater than those in thermocline. From the data obtained with the soaked bottles in situ, it was supposed that the decrease of vitamins in concentration during the daytime surpass consumption of vitamins by...

Proteomic signatures of the oyster metabolic response to herpesvirus OsHV-1 μVar infection ArchiMer
Corporeau, Charlotte; Tamayo, David; Pernet, Fabrice; Quere, Claudie; Madec, Stephanie.
Pacific oyster Crassostrea gigas were inoculated with OsHV-1 at low load (control) or high load (challenged) to better understand the pathogenesis of ostreid herpesvirus 1 (OsHV-1 μVar) and to determine which metabolic pathways might be affected during infection. Animals were sampled for proteomic analysis two days post-injection, at the same time as OsHV-1 initiated an intense replication phase in challenged oysters. Twenty-five abundant protein spots that showed a marked change in accumulated levels were identified using a two-dimensional electrophoresis (2-DE) proteomic approach. Overall, these proteins are involved in cytoskeleton organization, protein turnover, induction of stress signals, signalling pathways and energy metabolism. Challenged oysters...

Purine and pyrimidine nucleotide metabolism in Mollicutes Genet. Mol. Biol.
Bizarro,Cristiano Valim; Schuck,Desirée Cigaran.
Several mollicute genome projects are underway, offering unique opportunities to study genes and metabolic pathways on a genome-wide scale. Here, we have analyzed the conservation and diversity of purine and pyrimidine metabolism in mycoplasmas. An evaluation of discrepancies between genomic analysis and enzymatic data revealed interesting aspects about these organisms. We found important examples in which enzyme activity was reported without the annotation of a corresponding gene. An interesting example concerns phosphopentomutase. In Mollicutes, we have identified CDSs orthologous to sequences recently identified as new phosphopentomutases in archaeobacteria that are structurally related to phosphomannomutases. It is suggested that these sequences could...

Quality assessment of cryopreserved black-lip pearl oyster Pinctada margaritifera spermatozoa ArchiMer
Demoy-schneider, Marina; Schmitt, Nelly; Le Pennec, Gael; Suquet, Marc; Cosson, Jacky.
High quality of sperm is essential to a high fertilization rate, especially post- cryopreservation. Assessment of sperm integrity, motility and energy reserves before cryopreservation is necessary for selection of milt with optimal fertilizing potential. We describe the effect of cryopreservation on the quality of black-lip pearl oyster, Pinctada margaritifera var. cumingii sperm. Evaluated quality indices of fresh and frozen/thawed P. margaritifera spermatozoa, included morphology, ultrastructure and motility characteristics relative to the energy content (ATP) and its capacity to be sustained by mitochondrial respiration. Morphology and ultrastructure were quantitatively evaluated using images obtained by optical microscopy assisted by the Image J...

Reconstructing individual food and growth histories from biogenic carbonates ArchiMer
Pecquerie, Laure; Fablet, Ronan; De Pontual, Helene; Bonhommeau, Sylvain; Alunno-bruscia, Marianne; Petitgas, Pierre; Kooijman, Sebastiaan A. L. M..
Environmental conditions experienced by aquatic organisms are archived in biogenic carbonates such as fish otoliths, bivalve shells and coral skeletons. These calcified structures present an accretionary growth and variations in optical properties - color or opacity - that are used to reconstruct time. Full and reliable exploitation of the information extracted from these structures is, however, often limited as the metabolic processes that control their growth and their optical properties are poorly understood. Here, we propose a new modeling framework that couples both the growth of a biogenic carbonate and its optical properties with the metabolism of the organism. The model relies on well-tested properties of Dynamic Energy Budget (DEB) theory. It is...

For sustainable aquaculture, the removal of marine resource ingredients in fish diets is an important objective. While most studies focus on the replacement of fish oil by vegetable oil, little is known on the nutritional effects of presence (which corresponds to the control diet) or absence of dietary fish oil. We studied fatty acid composition of brush-border membranes and digestive enzyme activities of the intestine and measured the expression and activities of several enzymes involved in the hepatic intermediary metabolism of rainbow trout (Oncorhynchus mykiss) fed for 7 weeks with or without fish oil. The diets were pair-fed to ensure that fish fed either diet had comparable carbohydrate and protein intakes. Absence of fish oil significantly reduced...

Reduction du nitrate dans les sediments marins. Comparaison des activites mesurees in vitro et in situ ArchiMer
Esteves, J; Bonin, P; Blanc, F; Mille, G; Bertrand, J.
In vitro nitrate reduction in hydrocarbon-polluted marine sediments has been studied by flow-through system. By analysing the rate of nitrate comsumption as a function of nitrate concentration the authors determined the Michaelis-Menten kinetics parameters. (km = 330 mu M; vax = 2.0 mu M/g super(-1)/h super(-1). The authors found a value of 60 mu M/m super(-2)/h super(-1) for the rate of in vitro nitrate consumption derived from the above kinetic parameters and from the nitrate concentration in the sediment. In parallel a value of 67.4 mu M/m super(-2)/h super(-1) was found for the rate of nitrate consumption under in situ conditions. The great similarity between both results would favor the in vitro technique as a valid and more convenient one.

Paula,Edson Ferraz Evaristo de; Souza,Damaris Ferreira de; Monteiro,Alda Lúcia Gomes; Santana,Miguel Henrique de Almeida; Gilaverte,Susana; Rossi Junior,Paulo; Locatelli Dittrich,Rosangela.
The objectives of this study were to estimate the phenotypic correlations of residual feed intake (RFI) and gross feed efficiency (GFE) with hematological and metabolic blood profiles of lambs and to determine the differences for these traits in animals of different RFI classes. Twenty Ile de France male lambs, 115±8 days of age and 31.3±4.1 kg of body weight (means ± SD), were individually housed and their dry matter intake was measured over 65 days. They were weighed every 13 days to determine the average daily weight gain and two blood samples were collected at the last two weighings (at 07h30) for analysis of blood variables. The animals were divided into two classes: negative RFI (most efficient: <0.5 SD below the mean; n = 6) and positive RFI...

Role of bacteria in the gastral cavity of Anthozoa. ArchiMer
Herndl, G; Velimirov, B.
Bacterial density of the coelenteric fluid (CF) of some anthozoan species was determined in situ and under laboratory conditions. In all species examined bacterial density of the CF was higher in the gastral cavity than in ambient ranging from 10 super(5) up to 5 x 10 super(6) cells. ml super(-1). Incubation experiments with the CF indicate that a bacterial population within the coelenteron is efficiently controlled by the CF, favoring bacterial growth if bacterial densities are low in the CF and showing clearance activity at high bacterial densities. Feeding of pre-starved anthozoans caused a rapid response of coelenteric rod-shaped bacteria. Evidence is presented that coelenteric bacteria are digested periodically although they contribute 1% to the...

Seasonal and diurnal variability in carbon respiration, calcification and excretion rates of the abalone Haliotis tuberculata L. ArchiMer
Chapperon, Coraline; Clavier, Jacques; Dugué, Clément; Amice, Erwan; Le Goff, Marion; Roussel, Sabine.
Abalone (Haliotis spp.) are commercially important marine shellfish species worldwide. Knowledge about the physiology of abalone that impacts life-history traits is important for a better understanding of the biology of the species and the impact of stressful husbandry procedures at different seasons. The present study quantified the seasonal and diurnal variations in four physiological parameters of the European species Haliotis tuberculata, i.e. carbon aerial and aquatic respiration, calcification and excretion rates, and the effect of prolonged aerial exposure upon abalone aerial respiration. We also investigated the effect of individual size upon these physiological parameters. Aquatic respiration and calcification rates showed an allometric...

Short term variations in feeding and metabolism of Acartia tonsa (pelagic copepod) in the Berre lagoon (France) ArchiMer
Gaudy, R; Pagano, M; Cervetto, G; Saintjean, L; Verriopoulos, G; Beker, B.
Feeding, respiration, and ammonia excretion of Acartia tonsa were studied in a brackish lagoon, near Marseilles (Southern France). The experiments were performed during the same season in two successive years, using naturally occurring particles as food. Strong variations in both quantity and quality of food particles were recorded between the two series of experiments. The ingestion of particles according to their size was studied by means of the Coulter Counter technique. Females ingested more food than males. In both sexes, there was a tendency to select median to large particles, regardless of the shape of the particles-size spectrum. In 1990, the specific production rate, deduced from the energy budget, was equivalent to or greater than the observed...

Sodium chloride added to transport water and physiological responses of Matrinxã Brycon amazonicus (Teleost: Characidae) Acta Amazonica
Urbinati,Elisabeth Criscuolo; Carneiro,Paulo César Falanghe.
The addition of salt to the water has been used to mitigate stress and improve survival in fishes. This study investigated the effects of sodium chloride (0.0, 1.0, 3.0 and 6.0 g/l) on levels of plasma cortisol, glucose, tryacilglycerol, total protein, hematocrit, hemoglobin, erythrocyte number, liver glycogen and lipid, and muscle lipid in adult matrinxã(Brycon amazonicum) after a 4-h transport and during a 96-h recovery period. Fish were sampled before and after transport, and 24 and 96 h of the recovery period. Plasma cortisol was higher than initial condition immediately after transportation, except in fish transported in 3.0 and 6.0 g/l of salt. A similar pattern was observed for blood glucose but fish transported in water with 0.0, 1.0 and 3.0 g/l of...

Specific fatty acids as metabolic modulators in the dairy cow R. Bras. Zootec.
Pires,J.A.A.; Grummer,R.R..
This review summarizes recent developments on the utilization of specific fatty acids to modulate bovine energy metabolism, with emphasis on the periparturient dairy cow. A number of experiments have assessed the effects of polyunsaturated fatty acids on bovine hepatic energy metabolism using in vitro and in vivo models. Treatment of hepatocytes with specific fatty acids altered energy metabolism in vitro. For example, linolenic acid seemed to decrease hepatocyte triacylglycerol accumulation. This effect was confirmed in vivo, using parenteral infusions of emulsions derived from different fat sources to feed-restricted non-lactating cows. Additionally, polyunsaturated fatty acids can increase whole body response to insulin, potentially enhancing...

Sprint swimming performance of juvenile European sea bass ArchiMer
Nelson, J; Claireaux, Guy.
Despite its potential for determining animal success in a variety of ecologically important situations, very little is known about the sprint swimming performance of fishes. To evaluate whether selection on sprint swimming performance occurs in the wild, one must first demonstrate measurable and reproducible variation of sprint performance. In this study, we employ a computer-controlled laser detection system to study the sprint performance of 75 juvenile European sea bass Dicentrarchus labrax raised under uniform conditions in the laboratory. This percichthyid is a prized sport and food fish in Europe and similar ecologically to the North American striped bass Morone saxatilis. Individual European sea bass were subjected to endurance swimming performance...

Substitution of fish meal with plant protein sources and energy budget for white shrimp Litopenaeus vannamei (Boone, 1931) ArchiMer
Suarez, J. A.; Gaxiola, G.; Mendoza, R.; Cadavid, S.; Garcia, G.; Alanis, G.; Suarez, A.; Faillace, J.; Cuzon, Gerard.
Fish meal (FM) has long been considered an essential ingredient for the formulation of shrimp feeds. Until recently, 12%FM was considered a minimum amount, below which weight gain would become reduced due to a reduction in feed intake. In order to test this perceived minimum value, an experiment was conducted on a long-term basis with 4 replicates. Four practical isonitrogenous and isoenergetic diets were formulated for Litopenaeus vannamei juveniles. Survival, weight gain, feed conversion ratio (FCR) and protein efficiency ratio were determined for 0, 6, 10 and 15% FM diets and a reference diet. Shrimp survival ranged from 84 to 86.5%. Shrimp fed a diet with 0% FM showed lower average weight and SGR compared to the other treatments (P<0.05). FCR of...
